子痫前期患者胎盘组织TGF-β_1及MMP-9 mRNA的表达及其意义 被引量:2

摘要 目的研究转化生长因子β1(TGF-β1)及基质金属蛋白酶9(MMP-9)mRNA在胎盘组织中的表达情况及其与子痫前期发病的关系。方法用半定量逆转录聚合酶链反应(RT-PCR)技术对10例正常妊娠组和25例子痫前期组(包括11例轻度和14例重度)胎盘组织中TGF-β1、MMP-9 mRNA的表达水平进行检测。结果与正常妊娠组相比,重度子痫前期组胎盘组织中TGF-β1、mRNA表达量增加(P<0.05),MMP-9 mRNA的表达水平显着下降(P<0.01);轻度子痫前期组胎盘组织中TGF-β1、MMP-9与正常组相比无统计学差异(P>0.05)。结论TGF-β1、MMP-9的表达在基因转录水平就已经发生改变。TGF-β1通过下调MMP-9的表达而抑制滋养层细胞的侵入,这些浸润相关基因可能在子痫前期的发病中发挥着重要的作用。 Objective : To study the mRNA expression of transforming growth factor-beta1 (TGF-β1) and matrix metallopro-teinase 9 ( MMP-9) in placental tissue of pregnant women with preeclampsia and their relation to the pathogenic causes of preeclampsia. Methods : Semi -quantitative RT-PCR analysis was used to investicate the TGF-β1, MMP-9 mRNA expression levels in placental tissue of 10 cases of normal term pregnant women and 25 cases of women with preeclmpsia ( 11 cases of mild preeclampsia and 14 cases of severe preeclampsia). Results: Compared with the normal group, the mRNA expression level of TGF-β1 of severe preeclampsia group was higher ( P 〈 0. 05 ), the mRNA expression level of MMP-9 of severe preeclampsia group was significantly lower ( P 〈0. 01 ), there was no significance between the mild preeclampsia group and the normal group of the expression of TGF-β1 and MMP-9. Conclusions : The expression of TGF-β1 and MMP-9 was already changed at the level of transcription. By decreasing the expression of MMP-9, TGF-β1 inhibited the shallow invasion of trophoblast, these invasion related genes maybe play an important role in the pathogenic causes of preeclampsia.
