小鼠低温游泳脑缺血模型的建立及在注射液量效关系上的应用 被引量:2

A New Model of Brain Ischemic Model in Mice by Swimming in Cold Water And Application on Injections

摘要 目的:建立一种新的脑缺血小鼠模型,用标准的注射液验证模型的可靠性。方法:小鼠分成3组:生理盐水组,尼莫地平组(1.0mg/kg)和三七总皂苷组(120mg/kg)。静脉注射25min后,动物负重5%体重,在10~10.5气的水中游泳5min,取出,断头,记录断头的喘气持续时间。小鼠分别注射尼莫地平0.3、0.7、1.0、1.5mg/kg或三七总皂苷60、80、120、160mg/kg,同上法操作,探讨两种药物的量效关系。结果:和生理盐水组比较,两个药物能显著延长小鼠断头的喘气持续时间(P〈0.01)。尼莫地平的量效关系方程式为:Y=27.74919x+18.79446;三七总皂苷的量效关系方程式为:Y=0.13347x+20.51017,二者的量效关系均显著(P〈0.05,P〈0.01)。结论:新的方法能够用于评价药物的抗脑缺血作用。 Objective:To evaluate effect of a new model on brain ischemia of mice. Methods: Mice were divided into three groups : nimodipine group (1.0mg,/kg) ,physiologic saline(PS) group, and Panaxmoto Ginseng Sanponin(pns) group( 120mg/kg). After 25minutes of injection by vein, animals were set in a cold pool in 10 ~10.5℃for 5 minutes, loading 5% body weight. Then their heads were cut off and panting time was recorded. Mice were injected PNS in 60mg,/kg ,80mg,/kg, 120mg/kg,and 160mg/kg or nimodipine 0.3mg,/kg,0.7mg,/kg, 1.0mg,/kg,and 1.5mg,/kg respectively by vein. Both regression equations were studied in the model that the dose was independent variable and panting time was variance. Results: Comparing with PS group, the panting time of two groups was more lasted significantly (P 〈0.01 ). The regression equation of PNS is y =0. 13347x +20.51017 and the regression equation of nimodipine is y = 27.74919x + 18. 79446. The two dose - effect relationship in drugs appear significant linear ( P 〈 0.05 ,P 〈 0.01 ).. Conclusion : The new model can be applied to evaluating drug effect of anti - brain ischemia.
