HPLC法测定苦参及苦参汤有效部位中三叶豆紫檀苷含量 被引量:3

Quantitative determination of trifolirhizin in Sophora flavescens Ait. and the effective fraction of Kushen Decoction

摘要 目的建立测定苦参及苦参汤有效部位中三叶豆紫檀苷含量的高效液相色谱方法.方法 Phenomenex Luna C18(250 mm × 4.6 mm,5 μm)色谱柱,流动相为乙腈-磷酸水溶液[磷酸-水(2:530)](26:74),流速为1.0 mL/min,检测波长为310nm.结果三叶豆紫檀苷在0.161 6~0.8080μg范围内线性良好,相关系数r=0.9994.3组样品三叶豆紫檀苷平均回收率为98.97%、99.93%、97.94%,RSD为1.80%、1.84%、1.67%(n=3).结论本方法操作简便,分离效果好,灵敏度高,重复性好,可用于控制苦参及苦参汤有效部位质量. Objective To establish a method for the quantitative determination of the contents of trifolirhizin in Sophora flavescens Ait. and the effective fraction of Kushen Decoction by HPLC. Methods The HPLC procedure was used in the determination as follows: the chromatographic column was Phenomenex Luna Cts (250 mm× 4.6 mm, 5 tLm), the mobile phase was acetonitrile: phosphoric acid water solution [ H3PO3 - H2O ( 2 : 530 ) ] ( 26 : 74 ) with a flow rate of 1 mL/min, and the detection wavelength was 310 nm. Result The result showed that trifolirhizin had a good linear relationship in the range of 0. 1616 - 0.8080 tLg ( r = 0.9994 ). The average recoveries for trifolirhizin in three batches of samples were 98.97%, 99.93% and 97.94% respectively, RSDs were 1.80%, 1.84% and 1.67% respectively (n = 3 ). Conclusion The procedure was easy, sensitive with a good result and repeatability. It can be used to control the qualities of Sophora flavescens Ait. and the effective fraction of Kushen Decoction.
