链球菌制剂对NOD鼠胰岛瘤细胞株NIT生长影响的体外研究

Study of Group A Hemolytic Streptococci on Growth of NOD Mice Insulinoma Tumor Cell NIT

为了研究A群溶血性链球菌制剂对NOD鼠胰岛细胞瘤细胞株N IT体外生长的影响,利用不同浓度的制剂处理体外培养的NOD鼠胰岛细胞瘤细胞株N IT和正常小鼠胰岛β细胞,以不加药物的培养孔作为对照来对细胞进行形态学观察.以M TT法检测细胞活性,经A garose ge l电泳分析DNA ladder条带检测凋亡,结果表明:A群溶血性链球菌制剂能显著抑制N IT细胞增殖,而对正常胰岛β细胞没有影响,且该制剂有促N IT细胞凋亡的作用.

We try to investigate the role of Group A hemolytic streptococci on growth of NOD mice insulinma tumor cell NIT in vitro. Group A hemolytic streptococci diluted to different concentrations acted on NIT and mouse pancreatic islet β cells, the group without drugs as control researched cells through morphologic observation, Cell activity was tested by MTT method, cenn DNA fragmentation was analysised by electrophoresis in agarose gel to detect apoptosis. We found that Group A hemolytic streptococci can inhibit the proliferation and induce the apoptosis of NIT significantly, but it has not affect on normal islet cells. Group A hemolytic streptococci could lead NIT to apoptosis.
