HCV NS5b cDNA from two HCV infected individuals were sequenced with polymerase-chain-reation (PCR)-directed sequencing method. Compared with HCV-J, 91. 84% and 92. 36 % nucleotides homology, 91. 84% and 95. 79% homology with HC-C2,were found in HC114 and HC-C11 respectively. A three nucleotide deletion occurred in HC-114. As the generated profiles of sequencing ladders were clear, it indicated that PCR-directed sequencing method worked best in showing larger range variations as well as individual base variation.
