磷脂酶C-γ1在胚胎及新生小鼠脑组织的表达及意义 被引量:4

Expression and significance of phospholipase C-γ1 in brain tissue of embryonic and newborn mice

摘要 目的研究磷脂酶C-γ1在胚胎及新生小鼠脑组织的表达。方法制备胚胎及新生小鼠脑组织标本,进行石蜡包埋并切片,采用免疫细胞化学方法检测磷脂酶C-γ1的表达情况,并进一步利用Western blot技术对磷脂酶C-γ1的表达进行初步定量。结果免疫组织化学的结果表明,在胚胎脑组织中磷脂酶C-γ1的表达比较广泛,各种细胞均有表达,没有细胞特异性;而在出生后5d的脑组织,磷脂酶C-γ1特异地表达于大脑皮质层及神经核团内的神经元,周围的神经胶质细胞不表达。Western blot的结果表明,在胚胎各个组织(脑、四肢、心、肾、胃、胰等)中脑的表达最高,脑内的表达又随胚胎发育过程胚胎第11天(E11)、E14、E18、出生后5d逐渐降低。结论磷脂酶C-γ1相关信号通路对于小鼠胚胎脑的发育具有重要生物学作用,可能对于脑神经元发育、生长、分化等具有重要作用。 Objective To investigate the expression ofphospholipase C-γ1 (PLCγ1) in the brain tissue of embryonic and newborn mice. Methods The brain specimens from the embryonic and newborn mice were prepared by paraformadehyde-fixing, paraffin-imbedding and then being made into serial sections of 5 μm in thickness. The expression of PLCγ1 was examined by immunocytochemistry. Western blot was used to quantify the expression of PLCγ1. Results Immunocytochemistry showed that PLCγ1 was expressed extensively in the brain of embryonic mice without cell specific expression, while in the brain tissue of 5-day newborn mice, it was found specifically in neurons of cerebral cortex and nerve nuclei, not in neuroglial cells. Western blot revealed that PLCγ1 was expressed more in brain than any other tissues such as heart, kidney, stomach or pancreas, and so on. The expression in brain was in a decrement as the time went on (embryonic day 11, 14, 18, postnatal day 5). Conclusion PLCγ1 and the related signal pathway may play an important role in the development of mouse embryo brain and the growth and differentiation of neurons.
