核因子κB表达与胰腺癌转移及与MMP-2表达的关系 被引量:1

The relationship of expression of nuclear factor kappab gene and MMP-2mRNA and with lymph node metastasis of pancreatic cancer

摘要 目的探讨核因子κB(NF-κB,p 6 5)在胰腺癌(PC)中的表达与淋巴结转移及MMP-2表达的关系。方法免疫印迹法检测4 5例PC(淋巴结转移3 0例,非转移1 5例)和9例正常胰腺组织(NP)中p 6 5蛋白的表达;RT-PCR检测MMP-2mRNA的表达。结果p 6 5,MMP-2在4 5例PC组织中的阳性表达率分别为6 6.7%(3 0/4 5)和5 7.8%(2 6/4 5),在3 0例转移组织中的阳性表达率分别为8 3.3%(2 5/3 0)和7 3.3%(2 2/3 0),在非转移组织中的阳性表达率分别为3 3.3%(5/1 5)和26.7%(4/1 5),p 6 5与MMP-2表达呈明显正相关(r=0.7 4 3,P<0.0 5)。结论NF-κB异常表达与胰腺癌淋巴结转移有关,并与MMP-2正相关,NF-κB可能通过对下游基因表达的调控,参与了胰腺癌的转移过程。 Objective To study the relationship of expression of nuclear factor kappab gene and MMP-2 mRNA and with lymph node metastasis of pancreatic cancer. Methods The expression of NF-KB was investigated by Western blot in normal pancreatic(NP) samples of 9 cases and pancreatic cancer(PC) samples of 45 cases (lymph node metastasis 30 cases, non-lymph node metastasis 15 cases ). The expression of MMP-2mRNA was detected through semiquantitative reverse transcriptase polymerase chain reaction ( RT-PCR ) . Results Of 45 PC eases, the positive expression rate of p65 and MMP-2mRNA was 66.7% ( 30/45 ) and 57.7% (26/45) , respectively. Of 30 eases of lymph node metastasis, the positive expression rate of p65 and MMP-2mRNA was 83. 3% (25/30)and 73. 3 % (22/30) , respectively. Of 15 eases of non- lymph node metastasis, the positive expression rate of p65 and MMP-2mRNA is 33. 3% (5/15) and 26. 7% (4/15), respectively, and the expression of p65 gene was positively correlated with MMP-2mRNA expression( r = 0. 743, P 〈 0. 05 ). Conclusions The abnormal expression of NF-KB is closely associated with lymph node metastasis of PC. NF-kB is closely related to MMP-2, which indicated that NF-kB is involved in the lymph node metastasis of PC by mediating downstream gene expression.
