乳腺癌转移相关蛋白mRNA定量表达与临床相关性研究 被引量:1

Expression and Clinical Relationship of Correlated Protein mRNA with Breast Cancer Matastasis

摘要 目的探讨基质金属蛋白酶-2(MMP-2)和纤维粘连蛋白(Fibronectin,FN)mRNA表达量与乳腺癌转移的关系及临床意义。方法采用荧光定量RT-PCR方法检测30例乳腺原发癌和配对淋巴结转移癌以及116例乳腺癌的MMP-2和FN mRNA表达量,分析MMP-2和FN mRNA表达量与乳腺癌转移及临床病理因素相关性。结果30例淋巴结转移癌的MMP-2和FN mRNA表达量低于原发癌,组间比较差异有显著性(P<0.05)。肿瘤直径大、临床分期晚及淋巴结转移数目多的MMP-2和FNmRNA表达量低于肿瘤直径小、临床分期早及淋巴结转移数目少的乳腺癌。MMP-2和FN mRNA表达量与病理类型、组织学分级、雌、孕激素受体及Her-2表达无关。结论随着乳腺癌转移潜能的增加,MMP-2和FN在转录水平的表达下调。 Objective To discuss the clinical significance of matrix metalloproteinase-2 (MMP-2) and Fibronectin(FN) mRNA with breast cancer metastasis. Methods Detect MMP-2 and Fibronectin(FN) mRNA expression of 30 breast cancer, metastatic lymphnode and 116 clinical breast cancer samples by flurence-quantitative RT-PCR and analysis the relativity of MMP-2 and Fibronectin(FN) mRNA expression with clinical,pathology factors. Results MMP-2 and Fibronectin(FN) mRNA expression in 30 metastatic lymphnode is lower than in primary breast cancer(P〈0. 05). The mRNA expression of MMP-2 and Fibronectin(FN) in big tumor, late stage, more lymph nodes is lower than in others. The MMP-2 mRNA and Fibronectin(FN)expression is irrelated with tissue grade.pothologic type. ER.PR and Her-2. Conclusion MMP-2 mRNA and Fibronectin(FN)expression is down-regulated with tumor progressing, and related with the metastasis of breast cancer.
