FHIT在结直肠癌中的表达及其与Caspase-3、PCNA之间的关系 被引量:2

摘要 目的探讨FHIT蛋白、Caspase-3及PCNA在散发性结直肠癌(sporadic colorectal carcinoma,SCC)组织中的表达及其临床意义。方法采用免疫组织化学SP法检测92例SCC组织和28例正常结直肠组织中FHIT蛋白、Caspase-3和PC-NA的表达。结果(1)FHIT蛋白、Caspase-3、PCNA在正常结直肠组织及SCC组织中的阳性表达均有显著性差异(P<0.01)。(2)FHIT蛋白、PCNA表达与SCC组织学分级、Duke’s分期、淋巴结转移、5年生存率相关(P<0.01)。(3)Caspase-3的表达同SCC组织学分级、5年生存率相关(P<0.05)。(4)SCC组织中FHIT蛋白和Caspase-3两者表达呈明显正相关;FHIT蛋白和PCNA表达呈明显负相关(P<0.01)。(5)FHIT蛋白和Caspase-3,FHIT蛋白和PCNA联合表达与SCC的组织学分级、Duke’s分期、淋巴结转移及5年生存率均有关(P<0.05)。结论(1)FHIT蛋白、PCNA的表达与SCC的发生、恶性程度及预后有关;(2)Caspase-3的表达与SCC的发生、预后有关;(3)联合检测FHIT蛋白、Caspase-3及FHIT蛋白、PCNA可以用于筛查SCC的高危人群。 Objective To study the expression of FHIT (fragile histidine triad) protein.Caspase-3、PCNA in sporadic colorectal carcinoma (SCC) and their clinical significance. Methods Immunohistochemistry SP method was used to determine the expression of FHIT, Caspase-3 and PCNA in surgically resection specimens of 92 SCC and 28 normal colorectal tissues. Results (1) The positive different expression of FHIT protein.Caspase-3. PCNA in normal colorectal tissues and SCC was statistically significant(P〈0. 01). (2) Expression of FHIT protein and PCNA in SCC was closely correlated with the grading. Duke's stages.lymph node metastasis and 5-year survival(P〈0. 01 ). (3) Expression of Caspase-3 in SCC was closely correlated with grading and 5-year survival(P〈0. 05). (4) Significant positive correlation was in FHIT and Caspase-3;significant negative correlation was in FHIT and PCNA(P〈0. 01 ). (5) The combined expression of FHIT protein and Caspase-3; FHIT protein and PCNA in SCC were correlated with grade, Duke's stages, lymph node metastasis and 5-year survival rate(P〈0. 05). Conclusion (1)The expression of FHIT protein and PCNA in SCC was closely connected with ocurrence, progression and prognosis. (2)The expression of Caspase-3 in SCC was closely connected with occurrence and prognosis. ( 3 ) Improving specificity and positive expecting through combining examine FHIT, Caspase - 3 FHIT, PCNA in SCC.
