nm23-H1基因对口腔癌细胞骨架系统影响的实验研究

摘要 目的:探讨细胞骨架在nm23-H1基因抑制细胞转移行为和提高化疗敏感性的相关机制。方法:制备高纯度的nm23-H1真核表达质粒后,通过阳离子脂质介导的转染技术,完成转染方法的建立,利用激光共聚焦显微镜完成对细胞骨架的观察。结果:转染后细胞的微丝变得粗大,排列有序。结论:细胞骨架在改变肿瘤细胞黏附能力、运动能力、化疗敏感性方面起到了重要作用。 Objective:To study the mechanism of nm23-H1 for inhibiting the metastasis of Tca 8113 cell lines and improve the chemo-sensitivety to CDDP by observing the cytoskeleton.Methods:After transfecting nm23-H1 into Tca 8113 cell lines,the cytoskeleton was observed by laser confocal microscope.Results:The increased expression leve of nm23-H1 could avoid the destroy of cytoskeleton. Conclusion:Cytokseleton plays an important tale in the mechanism of inhibiting the metastasis and improving the chemo-sensitivety of CDDP.
