摘要
目的考察萘普生对大鼠Ⅱ型胶原性关节炎(CIA)的治疗作用,以及其对与类风湿性关节炎(RA)发病的相关免疫学指标的影响,并探讨其作用机制。方法用牛Ⅱ型胶原(CII)加完全佛氏佐剂(CFA)免疫Wistar大鼠建立胶原诱导性关节炎(CIA)模型,以足踝周长法测量动物足肿胀的变化并给出相应评分;ELISA法检测外周血中抗CⅡ抗体的水平、血清中TNFα和IL1的含量;以同位素法检测动物脾淋细胞在刀豆素A(ConA)的刺激下的增殖情况。结果萘普生可对CⅡ引起的大鼠足肿胀有明显的抑制作用;体外研究发现萘普生可使CIA大鼠过高的ConA增殖反应降低,同时对CIA大鼠血清中TNFα和IL-1释放有显著的抑制作用;并可显著抑制CIA大鼠体内抗CⅡ抗体的产生。结论萘普生对CIA大鼠具有治疗作用,该作用可能同时通过细胞和体液免疫调节实现的。
Objective To study the therapeutic effect of naproxen on type Ⅱ collagen arthritis(CIA) in rats,and its influence on the immunological indices related to the onset of rheumatoid arthritis (RA),and to explore its possible mechanism. Methods A CIA Wistar rat model was established using collagen II (CII) plus CFA. The ankle girths were measured to observe the changes of foot swelling and scored. The CII antibody level in the peripheral blood and the serum TNF a and IL1 concentrations were detected by ELISA method.The proliferation of splenic lymphocytes stimulated by concanavalin A (ConA) was detected by isotope method. Results Naproxen alleviated the rat foot swelling induced by CII, By in vitro study, it showed an effect on reducing the ConA proliferation in CIA rat, and inhibiting the release of serum TNF a and IL1. It also inhibited the production of CII antibody. Conclusion Naproxen has a therapeutic effect on CIA rats, and this effect may be realized by simultaneous cellular and humoral immunity regulations.
