摘要
目的:探讨天门冬提取物对糖尿病模型动物体质量、饮水量和血糖水平的影响。方法:实验于2005-04/07在甘肃政法学院公安分院实验中心完成。选用出生110dWistar大鼠50只(雌雄各半),动物正常饲养2d适应环境条件,从尾静脉采血,测定血糖水平。24h后,腹腔注射四氧嘧啶200mg/kg体质量,第5天采血,测定血糖水平。动物按血糖水平编号,随机数字表法分为5组,每组10只。天门冬5,10,20g/kg组[按5,10,20g/(kg·d)剂量灌胃天门冬提取物],降糖宁组(灌胃降糖宁0.3g/kg体质量),模型对照组(灌胃蒸馏水10mL/kg体质量)。灌胃5,10,20d,分别采血,用GlucoseKit(葡萄糖氧化酶法),721B型分光光度计测血糖水平。从实验开始,每天称量动物体质量,测定饮水量。计量资料差异比较采用Dunntt单因素方差分析。结果:Wistar大鼠50只均进入结果分析。①天门冬提取物对模型动物血糖水平的影响:分别给予模型动物天门冬提取物5,10,20g/kg体质量20d,血糖水平比模型组分别降低了69.3%,78.8%,92.4%(P<0.001)。②天门冬提取物对模型动物日饮水量和体质量的影响:分别给予模型动物天门冬提取物5,10,20g/kg体质量20d,体质量比模型组分别增加了16.2%,22.2%,27.9%(P<0.05~0.001);日饮水量比模型组分别减少了53.5%,53.5%,58.5%(P<0.001),给药组间差异无显著性意义。结论:天门冬提取物具有降低四氧嘧啶糖尿病模型动物血糖,减少饮水量和增加体质量的作用。
AIM: To study the effects of lucid asparagus extract on body mass, volume of drinking water and level of blood glucose in diabetic models.
METHODS: The experiment was performed at the Experimental Centre of Public Security, Gansu Political Science and Law Institute from April to July 2005. Totally 50 Wistar rats aged 110 days, of male and female (half and half), were selected. After 2-day normal feed, the animals were adapted to the environment. Blood was isolated from vena eaudalis. Level of blood glucose was determined. After 24 hours, Wistar rats were given 200 mg/kg body mass of alloxan (intraperitoneal injection) and blood samples were taken from tail blood vessel at day 5 after administering alloxan. The level of blood sugar was determined. Animals were numbered based on level of blood sugar and randomly assigned into 5 groups with 10 in each group. The rats in the lucid asparagus extract 5, 10, 20 g/kg group were treated with 5, 10, 20 g/kg lucid asparagus extract daily by gastric perfusion. The rats in the Jiangtangning group were treated with 0.3 g/kg Jiangtangning by gastric perfusion. The rats in the model control group were treated with 10 mL/kg distilled water by gastric perfusion. After gastric perfusion for 5, 10 and 20 days, blood was collected, respectively. Level of blood sugar was detected with Glucose Kit method and 721B speetrophotometer. From the beginning, body mass and volume of drinking waster were determined daily. Difference of measurement data was compared with Dunntt single factor variance analysis.
RESULTS: A total of 50 rats were involved in the result analysis. ①Effect of lucid asparagus extract on level of blood sugar of model animals: After the rats were treated with lucid asparagus extract 5, 10, 20 g/kg for 20 day, level of blood glucose decreased by 69.3% ,78.8% ,92.4%(P 〈 0.001 ) as compared with the model group. ②Effect of lucid asparagus extract on volume of drinking water and body mass in a day: After the rats were treated with lucid asparagus extract 5, 10, 20g/kg for 20 day, body mass increased by 16.2%,22.2%,27.9%(P 〈 0.05-0.001) as compared with the model group. Daily volume of drinking water decreased by 53.5% ,53.5%, 58.5% (P 〈 0.001 ) as compared with the model group. There was no significant difference between administration groups.
CONCLUSION: Lucid asparagus extract has the effect of reducing blood sugar, decreasing volume of drinking water and increasing body mass in alloxan diabetic model animals.
