治疗脑血管疾病中成药物金属元素的模糊分类(英文) 被引量:1

Vague categorization of the metal elements in Chinese patent medicines for treating cerebrovascular diseases

摘要 背景:微量元素与脑血管疾病的病因和发病机制密切相关,很多微量元素在维持心脑血管功能及结构中起着有益的作用,人体内所需的微量元素在中成药中均存在,不同的中成药含有不同的微量元素。目的:对10种临床常用治疗脑血管疾病的中成药物中金属元素进行模糊分类,找到药物间微量和宏量元素含量的相似性,为研究该类药物中微量元素的作用及与治疗脑血管疾病的关系提供依据。设计:本实验对10种中成药物粉碎准确称质量后,采用HNO3-HClO4湿法消化,用原子吸收分光光度计测定其金属元素的含量,并运用数学矩阵,对数据进行模糊分类,以期找到药物间相似点。单位:吉林医药学院化学教研室,吉林医药学院附属医院神经内科。材料:脑塞通片、强力天麻杜仲丸、通塞脉片、脉通胶囊、通脉宁心冲剂、复方银杏胶丸、大活络丹、中风回春片、消栓通络胶囊和地奥脂必妥,共10种,均为市售。试剂:Ca、Mg、Cu、Cr标准储备液均为国家标准溶液(NCS),浓度均为1.000g/L(国家钢铁材料测试中心);Mn标准储备液均为国家二级标准溶液,浓度为500mg/L(中国环境监测总站);使用液Ca、Mg、Cu、Cr浓度均为50mg/L,Mn为20mg/L,优级纯HNO3、HClO4均为优级纯,实验用水为去离子水。方法:于2001-03/06在吉林医药学院化学教研室选择上述临床上常用的治疗脑血管疾病中成药物10种,烘干研碎,充分混匀,用分析天平准确称取样品1.0000g,每种平行取3份,用原子吸收分光光度法测定其宏量元素钠、钙、镁、钾和微量元素铁、镍、锰、铬、铜、锌10种无机元素的含量及铜/锌、钙/镁比值,运用模糊分类的方法研究其药物的相关性。主要观察指标:通过药物中微量元素含量的测定,观察不同药物中微量元素的含量差别,并在数据聚类分析中找到其相似点。结果:①测定结构显示,10种药物中宏量和微量元素丰富,但不同药物中含量有所差别,宏量元素含量较多的药物是脑塞通片、强力天麻杜仲丸、通塞脉片和大活络丹;微量元素含量较丰富的是通塞脉片和脉通。②将每个样品中10种金属元素含量数据、铜/锌、钙/镁比值作为指标,进行统计指标的标准化处理,并用最大树的方法进行聚类分析,在r=0.65时,有5种药物聚类相似,相似的药物有脑塞通片、强力天麻杜仲丸、通塞脉片、脉通和消栓通络胶囊。③当r=0.85时,有3种药物聚类相似,分别是强力天麻杜仲丸、通塞脉片和脉通;r=0.95时,有两种药物微量元素聚类分析极为相似,分别是通塞脉片和脉通。从药效上看,通塞脉片和脉通均具有增加血流量,改善血流作用,在治疗脑血管疾病中有很多相同点。结论:药物在治疗过程中微量元素应有其相似的作用,聚类分析相关系数大的中成药在治疗脑血管疾病中疗效相似程度也很大。相关系数较小的药物在治疗过程中,药物的疗效相似点也少。 BACKGROUND: Epidemiological studies indicated that trace elements were closely correlated with the causes and pathogenesis of eerebrovascular disease, many trace elements play a beneficial role in maintaining the eerebrovascular and cardiovascular functions and structures, the trace elements needed in human body all exist in Chinese patent medicines, and different Chinese patent medicines contain different trace elements. OBJECTIVE: To make vague category of metal elements in 10 kinds of Chinese patent medicines commonly used to treat cerebrovaseular diseases in clinic, and find the similarity of the contents of trace elements and maeroelements among the medicines, so as to provide evidence for researching the role of trace elements in the medicines and the correlation with the treatment of eerebrovaseular disease. DESIGN: Ten kinds of Chinese patent medicines were powdered and accurately weighed, digested wit the HNO3-HClO4 wet method, the contents of metal elements were determined with atomic absorption spectrometry, mathematic matrix was applied in the vague,category of the data so as to find the similarities among the medicines. SETTINGS: Department of Chemistry, Jilin Medical College; Department of Neurology, Affiliated Hospital of Jilin Medical College. MATERIALS: The medicines were Naosetong tablet, Qiangli Tianma Duzhong Wan, Tongsemai tablet, Maitong capsule, Tongmai Ningxin Chongji, Fufang Yinxing Jiaowan, Dahuoluo Dan, Zhongfeng Huichun tablet, Xiaoshuan Tongluo capsule and Di'ao Zhibituo, all of which could be bought on the market. Reagents: The standard reserving solution for Ca, Mg, Cu and Crwas the national criteria solution (NCS) with the concentration of 1.000 g/L (State Test Center of Ferrous Materials), that for Mn was the secondary NCS with the concentration of 500 mg/L (China General Station for Environment Monitoring). The concentration was 50 mg/L for Ca, Mg, Cu and Cr, 20 mg/L for Mn, HNO3 and HClO4 were both in excellent purity, and the water for the experiment was deionized water. METHODS: From March to June in 2001, 10 kinds of Chinese patent medicines commonly used to treat eerebrovaseular diseases in clinic were selected in the Department of Chemistry, Jilin Medical College, dried and powdered, completely mixed, 1.000 0 g was weighed accurately with analytical balance, 3 portions were used for each kind of sample. The atomic absorption spectrometry was used to determine the contents of maeroelements (Na, Ca, Mg, K) and trace elements (Fe, Ni, Mn, Cr, Cu, Zn), the ratios of Cu/Zn and Ca/Mg were calculated, the method of vague category was applied to study the correlations of the medicines. MAIN OUTCOME MEASURES: The differences of the contents of trace elements in different medicines were observed by determining their contents, and the similarities were found in the clustering analysis of the data. RESULTS: ① The contents of the trace elements and maeroelements were rich in the 10 medicines, but they were different in different medicines. There were more maeroelements in Naosetong tablet, Qiangli Tianma Duzhong Wan, Tongsemai tablet and Dahuoluo Dan; The trace elements were more in Tongsemai tablet and Maitong capsule. ② The contents of 10 kinds of metal elements, Cu/Zn ratio and Ca/Mg ratio of each sample were taken as the indexes for standardized processing, and the method of maximal tree was applied for the clustering analysis. When r=0.55, five medicines had similar cluster, which were Naosetong tablet, Qiangli Tianma Duzhong Wan, Tongsemai tablet, Maitong capsule, Dahuoluo Dan and Xiaoshuan Tongluo capsule. ③ When r=0.85, three medicines had similar cluster, which were Qiangli Tianma Duzhong Wan, Tongsemai tablet and Maitong capsule. When r=0.95, the clustering analysis of trace elements of two medicines were extremely similar, which were Tongsemai tablet and Maitong capsule. Both Tongsemai tablet and Maitong capsule had the efficacies of increasing blood flow, ameliorate blood stream, and they had many similarities in treating cerebrovascular disease. CONCLUSION: Trace elements should have the similar actions in the process of drug treatment, the Chinese patent medicines with greater correlation coefficient in the clustering analysis have greater similarities in the efficacy on cerebrovascular disease, and those with smaller correlation coefficient also have fewer similarities in the efficacy.
