摘要
用共价连接方式将专一切割苜蓿夜蛾核多角体病毒多角体蛋白mRNA片段的锤头状ribozyme(hammerheadribozyme)固定在SepharoseCL-4B上,ribozyme与载体之间有19个原子的距离,研究并比较了该固定化ribozyme与游离(非固定化)ribozyme的底物专一性、催化活性、最适反应温度以及热和保存稳定性。讨论了固定化ribozyme与固定化酶在性质上的相似性以及固定化ribozyme研究在理论和应用上的可能意义。
A hammerhead ribozyme directed against polyhedrin gene of Autographa californica nuclear polyhedrosis virus was immobilized covalently on the carrier Sepharose CL-4B through amide bond. The distance between carrier and ribozyme is 19 atoms apart which is believed to be good enough for the ribozyme to react with substrate targets. The immobilized ribozyme showes same specificity as the free ribozyme. Its properties such as the catalytic efficiency,optimum temperature,thermal stability and storage stability are somewhat different from that of the free ribozyme.
