摘要 目的:建立双丹颗粒中丹酚酸B和芍药苷的含量测定方法。方法:采用高效液相色谱法,Zorbax SB-C18色谱柱,流动相A0.05%的磷酸水溶液,B含0.05%磷酸的4%乙腈甲醇溶液,梯度洗脱,流速1mL·min^-1,柱温25℃,检测波长230nm。结果:丹酚酸B在0.710~22.7μg(r=0.9999),芍药苷在0.103~3.30μg(r=0.9999)有良好的线性关系,测定了3个批次双丹颗粒中丹酚酸B和芍药苷的含量,其中丹酚酸B的含量范围为6.46~11.6mg·g^-1,芍药苷的含量范围为1.44~1.78mg·g^-1。结论:该方法准确度好、重复性好、稳定可靠,可以作为双丹颗粒质量控制的方法。 Objective: To develop a method for the determination of salvianolic acid B and paeoniflorin in Shuangdan granules. Method: The chromatographic separation was performed on a Zorbax SB C18 column with a linear gradient elution of phosphoric acid-water (0.05:100) and phosphoric acid-acetonitrile-methanol (0.05:4:96) was used. The flow rate was 1 mL·min^-1 and column temperature was set at 25 ℃. The UV detection wavelength was set at 230 nm. Result: The linear ranger was of 0.710-22.7 μg for salvianolic acid B and 0.103-3.30 μg for paconiflorin. Three hatches of Shuangdan granules were analyzed, and the content of salvianolic acid B ranged from 6.46- 11.6 mg·g^-1 and paconiflorin ranged from 1.44-1.78 mg·g^-1. Conclusion: The method was accurate, reproducible, reliable, and could be used for quantitative control of Shuangdan granules.
