摘要
目的:研究抑癌基因RASSF1A在乳癌组织中的表达情况及与RASSF1A基因启动子甲基化的关系。方法:运用RT-PCR方法检测40例乳癌组织和6例乳腺良性肿瘤和5例正常乳腺组织中RASSF1A的表达,运用甲基化特异PCR方法检测这些组织中RASSF1A基因启动子的甲基化情况。结果:RASSF1A基因在40例乳腺癌组织中8例(20%)存在表达缺失,差异无统计学意义(P>0.05)。但其表达量明显低于正常乳腺组织(P<0.05)。乳腺癌组织的RASSF1A基因启动子甲基化频率(65%)明显高于正常乳腺组织(20%)和乳腺良性肿瘤(17%)(P<0.05)。结论:乳腺癌组织中RASSF1A基因启动子的高甲基化是引起RASSF1A基因表达下调的重要原因,在肿瘤的发生和发展中起重要的调控作用。
Objective:To study the expression of the tumor suppressor gene RASSFIA in breast cancer, and the correlation between the expression and the promoter bypermethylation of RASSFIA. Methods:RT - PCR was used to detect the mRNA expressions of RASSFIA in 40 cases of breast cancer,6 cases of benign carcinoma and 5 cases of normal breast tissue . We examined the promoter methylation status of RASSFIA by using a methylation - specific PCR. Results : RASSFIA expression was not detected in 8 of the 40 cases of breast cancer without a statistical significance in the difference(P 〉 0. 05 ). But a decrease in expression of RASSFIA in breast cancer was of a statistical significance( P 〈 0. 05 ). The frequence of the promoter hypermethylation of RASSFIA in breast cancer(65% ) was significantly higher than in normal breast tissue(20% )and benign carcinoma ( 17% ). Conclusion:The promoter hypermethylation of RASSFIA in human breast cancer plays an important role in the low expression of RASSFIA,and is involved in the genesis and development of breast cancer.
