摘要
采用RAPD方法对国内十种栽培苦荞麦的基因型进行分析,建立了它们的指纹图谱。从60个随机引物中筛选出的6个有效引物共产生44条DNA片段,大小分布在0.1-2.0Kb之间,其中18个条带具有遗传多态性,约占总数的40.90%,平均每个引物扩增的DNA带数为7.33条。十种栽培苦荞麦的遗传相似性分析表明,各基因型之间的J相似性系数分布在0.8525~0.9846之间,平均相似性系数为0.9185。应用NTSYS-pc软件进行聚类,将聚类结果转化为十种栽培苦荞麦基因型之间的遗传关系树型图。十种栽培苦荞麦基因型聚为A、B、C三类,A类包括六苦1号、六苦2号、定98-1、西农9909、黔威3号和九江苦荞6个栽培种,B类包括六盘水野生苦荞、昭苦1号和滇宁1号3个栽培种,C类仅为六苦3号1个栽培种。
The fingerprints of 10 species genotypes of Tartary Buckwhea were generated using RAPD method. Six primers were screened from 60 ten-bp arbitrary Primers, and a total of 44 DNA fragments were amplified ranging from 0. 10 - 2.0 Kb, among them 18 (40. 90% ) bands were polymorphic. The average number of DNA band produced by each Primer was 7.33. The results of genetic similarity analysis for 10 species genotypes of Tartary Buckwhea Showed that the J' s coefficient ranged from 0. 852 5 - 0. 984 6, and the average J' s coefficient was 0.918 5. A DNA molecular dendrogram was established for 10 species genotypes of Tartary Buckwhea based on cluster analysis with the help of NTYSY-pc. It divided the 10 species genotypes of Tartary Buckwhea into three groups: group A,group B and group C. Group A consisted of No. 1 ,No. 2,No. 9,No. 10, No. 5 and No. 8, amount to 6 genotypes. Group B consisted of No. 4, No. 6 and No. 7, amount to 3genotypes. Group C included No. 3 only 1 genotypes.
