摘要
目的对脂质体流感疫苗体液免疫和细胞免疫效果进行评价。方法血凝抑制法(HI)和ELISA法检测特异性抗体;MTT法检测小鼠淋巴细胞增殖情况;流式细胞仪测小鼠T淋巴细胞表面标记;ELISA法检测小鼠肺内溶物中细胞因子含量。结果脂质体流感抗原可激发更强的体液免疫;脾细胞增殖效率,CD4水平和CD4/CD8比例,以及细胞因子(IL-4、IFN-γ)水平均有显著的增强作用。结论脂质体流感疫苗能够刺激机体产生较高的体液免疫,并且能够激发机体产生细胞免疫。
Objective To produce liposome-encapsulated influenza split vaccine and evaluate its humoral and cellular immune effect. Methods Produce liposome-encapsulated influenza split vaccine, to measure special IgG with HI and ELISA tests;using MTT to measure the spleen cell proliferation in vitro;to determine the levels of CD4 and CD8 T-cell and the CD4/CD8 using flow cytometry; to quantify the levels of IL-4 and IFN-γ in lung homogenates by ELISA kit. Results the liposome-encapsulated influenza split vaccine elicited high HI titer, stimulate spleen cell proliferation, increase cell subsets like CD4^+ and CD4/CD8 and to trigger cytokine (IL-4 and IFN-γ) production. Condusion liposome-encapsulated influenza split vaccine can stimulate humoral and cellular responses efficiently.
