HCMV感染致内皮细胞粘附分子表达的变化 被引量:5

摘要 目的探讨HCMV感染对体外培养内皮细胞粘附分子表达的影响。方法应用双抗体夹心酶联免疫分析法及细胞ELISA方法检测体外培养人脐静脉血管内皮细胞在感染HCMV后,血管内皮细胞粘附因子(VCAM-1)及可溶性细胞间粘附因子1(sICAM-1)表达的变化。结果感染组细胞培养液sICAM-1 24小时和对照组相比明显升高达10.7±2.4μg/L,而正常对照为5.7±3.1μg/L,而后48小时、72小时均高于相应对照组(P<0.05)。VCAM-1在感染12小时开始表达升高,48小时和72小时和对照组相比明显增高(P<0.05)。结论体外情况下HCMV感染可上调内皮细胞粘附分子表达,这可能在HCMV致动脉粥样硬化的病理过程中发挥重要作用。 Objective To study Effects of human cytomegalovrius infection on the Expression of cellular Adhesion Molecules in cultured Endothelial cells.Methods After ECs were infected by HCMV AD169 strain,the vascular cell adhesion molecule-1 was determined by cell enzyme-linked immunosorbent assay,and the soulable intercelhr adhesion molecule-1 sICAM-1 was determined by ELISA.Results At different infection time (24 h,48 h,72 h) , ELISA showed that the value of sICAM-1 were 10.7±2.4μg/L, 16.6±5.4μg/L, 17.7± 6.6 μg/L, which were higher than that of control group. CeI1-ELISA also showed that the level of VCAM-1 were significantly higher than that of control group. Conclution HCMV infection may up-regulate the expression of cellular Adhesion Molecules of ECs, which may play an important role in the pathogenesis of atherosclerosis.
