银芩胶囊抗流感病毒的活性

Anti-influenza virus activity of Yinqin capsules

目的研究银芩胶囊在体内、外的抗流感病毒活性。方法采用噻唑蓝(MTT)比色法检测银芩胶囊对体外模型狗肾细胞(MDCK细胞)的毒性浓度;采用细胞病变效应(CPE)观察法测定银芩胶囊体外抑制流感病毒有效浓度;流感病毒感染动物模型为昆明种小鼠,口服给予银芩胶囊剂量分别为每日1.0、0.3和0.1g/kg,分2次给予,共给药6d,通过观察流感病毒FM1感染小鼠死亡率、生命延长情况及肺病变程度指标判定其抗流感病毒作用。结果银芩胶囊在1.25mg/mL浓度下对MDCK细胞无毒性,在0.6mg/mL浓度下能有效抑制流感A3病毒CPE产生;每日口服1.0、0.3和0.1g/kg银芩胶囊,对流感病毒FM1感染小鼠的死亡保护率分别为40%、20%和5%,生命延长率分别为51.1%、29.1%和23.4%,减轻小鼠肺病变分别为64.3%、56.1%和34.6%。结论银芩胶囊在体内、外均有良好的抗流感病毒活性。

Purpose To study the antiviral activity of Yinqin capsules against influenza virus in vitro and in vivo. Methods The cell culture technique was used in MDCK cells to get cell viability of different concentration Yinqin capsules by MTT assay. And virus inhibitory rate was determined by Cytopathic effect assay. The in vivo anti-virus activity was performed on Kunming mice divided to receive oral Yinqin capsules in twice daily dose of 1.0, 0.3 and 0.1 g/kg for 6 days. The mean survival days and the pulmonary pathological lesions of the infected mice were observed to evaluate the therapeutic efficacy of Yinqin capsules. Results Yinqin capsules showed no toxicity to MDCK cells at 1.25 mg/ mL, and could effectively inhibit influenza A3 virus at 0.6 mg/mL in vitro. It indicated that 1.0, 0.3 and 0.1 g/kg per day dosage medicine orally given bid for 6 days could reduce the death rate of the infected mice by 40% ,20% and 5% respectively, and could prolong the mean survival days by 51.1%, 29.1% and 23.4% respectively, and the pulmonary pathological lesions were reduced by 64.3%, 56. 1% and 34. 6 % respectively. Conclusions Yinqin capsules is an effective anti-influenza virus compound in vitro and in vivo.
