摘要
从感病的豆天蛾幼虫中分离和纯化了一种新的豆天蛾核型多角体病毒(CbNPV)。为进一步了解该病毒的分子生物学特征,提取了基因组DNA,采用Shotgun方法构建了病毒DNA的文库,对其中一些克隆分析发现一长度为465bp的读码框,编码154个氨基酸,在GenBank中没有发现同源性极高的核苷酸序列。但该基因的编码产物与sod基因同源性较高,与NPVs及GVs的同源性分别为67%-82%和54%-59%,并且更接近于Ⅱ类NPV。在CbNPVsod基因的氨基酸序列中一些与SOD活性相关的住点及维持空间构型二硫键的位点均保守。CbNPV的全基因组序列有待进一步解析。
A novel baculovirus, Clanis bilineata nucleopolyhedrovirus(CbNPV) was isolated and purified from Clanis bilineata. The genomic DNA of CbNPV was extracted, and a DNA fragments library was constructed by Shotgun. Sequence analysis indicated that an ORF of superoxide dismutase gene contains 565 base pairs and encodes 154 amino acids. The result of amino acid sequence homology analysis showed that CbNPV SOD has 67% - 82% and 55% - 59% identities with that of NPVs and GVs, respectively. It had closer relationship with Group II NPVs. There were some function domains conserved in SOD amino acid sequence.
