摘要
根据梨S基因高度保守区C1和C3区,设计1对引物P1和P2,对梨品种的基因组DNA进行S基因特异扩增、克隆、测序,并在GenBank中BLAST比较,确定S基因特异性片段,对京白梨等6个供试自交不亲和品种的S基因型比对结果为:白梨中的‘库尔勒香梨’为S21S28,‘苹果梨’为S17S19;砂梨中的‘台湾蜜梨’为S11S22;西洋梨中的‘葫芦梨’为SaSb;秋子梨中的‘京白’为S16S30,‘早梨18’为S4S28。其中S28和S30为首次登录的新S基因,在GenBank的登录号分别为AY562394(库尔勒香梨)和AY876945(京白)。
Partial S-RNase genes of pear were amplified and cloned by using a pair of primers (P1 and P2) based on the conserved regions C1 and C3 of pear S-RNase structure in six cultivars of unknown S-geontypes. The PCR products were sequenced and compared in GenBank. The S-genotypes of the six tested selfincompatible cuhivars were identified as follows : ‘Kuerle Xiangli' (Pyrus bretschneideri Rehd. ) was S21S28, ‘Pingguoli' (P. bretschneideri Rehd. ) was S17S19, ‘Taiwan Mili' (P. pyrifolia Nakai) was S11S22, ‘Hululi' ( P. communis L. ) was SaSb, ‘Jingbai' ( P. ussriensis Max. ) was S16S30‘Zaoli 18' ( P. ussriensis Max. ) was S4S28. S28-RNase and S30-RNase were deposited in GenBank under the accession numbers of AY562394 and AY876945.
