原发性肝癌基质金属蛋白酶-9的表达与肿瘤侵袭关系

Expression of MMP-9 mRNA in human hepatocellular carcinoma

摘要 目的探讨基质金属蛋白酶9(MMP9)在原发性肝癌中的表达及其与肝癌侵袭的关系。方法采用半定量逆转录聚合酶链反应(RTPCR)检测比较28例肝癌组织和癌旁组织中MMP9基因的表达水平。结果MMP9mRNA在肝癌组织中存在过度表达,显著高于癌旁组织(P<0.01);伴有门静脉癌栓的肝癌组织中MMP9mRNA表达值显著高于不伴有门静脉癌栓者(P<0.01),MMP9mRNA与术前AFP、肿瘤大小、TNM分期等无关。结论MMP9基因的过度表达与肝癌的侵袭密切相关,可以作为预后判断指标。 Objective To investigate the expression of MMP-9 genes in primary hepatoeeUular carcinoma (HCC). Methods RT-PCR was used to detect MMP-9 mRNA in surgically extracted specimens from 28 patients with HCC. Results The expression of MMP-9 mRNA was significantly higher in tumor than in corresponding nontumoral tissue( P 〈 0.01 ). And there was no significant correlation between the expression level and preoperative AFP level,tumor size, TNM staging, and portal vein tumor thrombus( P 〉 0.05). Meanwhile, the expression of MMP-9 mRNA in HCC was significantly correlated to the existence of portal vein tumor thrombus( P〈 0.01 ). Conclusion The overexpression of MMP-9 is closely correlated to recurrence and metastasis of HCC. MMP-9 expression level can be used as a predictor for-prognosis of HCC.
