蛇毒类凝血酶的纯化及鉴定

The Purification and Identification of Thrombin-like Enzyme from Guangxi Agkistrodon Acutus Venom

采用Source 30Q及Phenyl sepharose HP柱层析进行纯化,并用体外MTT实验研究了蛇毒类凝血酶抗肿瘤转移作用。结果得到SDS-PAGE条带均一、分子量为27.6 ku的类凝血酶I11-H2,体外MTT实验证明,I11-H2具有较理想的抗肿瘤转移作用。

The snake venom thrombin-like enzyme（SVTLE） was purified by successive chromatography on Source 30Q and Phenyl sepharose HP columns from Agkistrodon acutus Venom in Guangxi. And the restraining effects of SVTLE on tumor metastasis were researched by MTT in vitro. The purified SVTLE named I11-H2 was only one band with 27. 6 ku by SDS-PAGE. And the purified SVTLE（I11-H2 ） could restrain tumor metastasis by MTT.
