IL-8对心肌缺血再灌注组织中P53 mRNA和Bcl-2 mRNA的影响 被引量:7

摘要 目的探讨IL-8对心肌缺血再灌注(M IR)组织中P53、Bc l-2基因的影响。方法建立大鼠心肌缺血再灌注模型,分为对照组和实验组,缺血30m in后,再灌注2、4、8、12、24、48h,各时间点分别处死动物,取血备用。采用双抗体夹心酶联免疫吸附法(ELISA),原位杂交法,原位末端标记染色等,检测IL-8,P53 mRNA,Bc l-2 mRNA和细胞凋亡。结果从M IR的时间窗来看,IL-8在IR 2h即开始升高,12h达高峰。P53在4h开始升高,24h达高峰。Bc l-2在8h开始升高一直持续到48h达高峰。IL-8早于P53表达,Bc l-2晚于P53表达,心肌细胞凋亡和P53 mRNA表达时间相一致。结论IL-8介导心肌凋亡在继发的心肌局部功能的损害中扮演了一个重要角色,并可作为早期诊断心肌缺血再灌注损伤新的标记物。
