延胡索中生物碱成分的分离与反相高效液相色谱法含量测定 被引量:7

摘要 目的建立分离、测定延胡索中四种生物碱成分包括去氢紫堇碱、巴马汀、黄连碱、非洲防己碱含量的方法。方法采用10%氨水湿润药材粉末并用氯仿回流提取总生物碱,高速逆流色谱法分离制备各生物碱成分;利用反相高效液相色谱法测定各生物碱成分的含量。以乙腈-0.03 mol/L磷酸二氢钠(32∶68,每升流动相加入1.670 g十二烷基磺酸钠)为流动相,检测波长345 nm,流速0.6 m l/m in。结果各生物碱成分在测定范围内线性关系良好。结论该方法准确、快速,适用于延胡索药材的质量控制。 The dried powder of tubers of Corydalis yanhusuo was soaked in 10% ammonia solution and extracted with chloroform under refluxlng for three times to give the total alkaloids. Four quaternary alkaloids including dehydrocorydaline, palmatine, coptisine and columbamine were isolated from the total alkaloids by semi - prepaxative high speed counter - current chromatography and a RP - HPLC method was developed to determine their contents in the plant. It was separated on a Shim - Pack CLC ODS (M) C18(5μm,4.1 mm ×250 mm) column with acetonitrile -0.03 mol/L potassium dihydrogen phosphate (32: 68, v/v, added 1. 670 g of sodium dodecyl sulphonate to 1 000 ml mobile phase) as the mobile phase. The UV detector was set at 345nm and the flow rate was 0.6 ml/min. Good linear calibration curves was obtained in the injection ranges. This method was proved to be accurate and quick, and can be selected to control the quality of the medicinal materials.
