近交系小鼠微卫星位点的遗传标记多态性研究 被引量:1

摘要 目的研究近交系小鼠微卫星位点的遗传标记多态性及其意义。方法随机选取不同染色体的5个微卫星位点,应用PCR技术对BALB/C、豫医无毛小鼠、CBA、C57、DBA/2、C3H/HeJ、FVB/NJ及昆明小鼠共八种小鼠的DNA多态性进行研究。结果5对引物在近交系小鼠各基因座上均出现一条清晰的带,D16m it4、D3m it17、D3m it18具有多态性,D4m it2、D2Nds3不具有多态性。结论筛选出的引物D16m it4、D3m it17、D3m it18可用于检测近交系小鼠的品系来源和遗传背景,且所检测的小鼠符合近交要求。 Aim: To study the genetic marker polymorphisms and role of microsatellite of inbred strain mice. Methods : Five microsatellites DNA loci on different chromosomes in eight kinds of strain mice including BALB/C .Yuyi hairless mice.CBA.C57 .DBA/2 .C3H/HeJ.FVB/NJ and Kunming mice were investigated by PCR analysis. Results : all these mice tested display single allelic gene band with five pairs of primers, Polymorphism were found at D16mit4.D3mit17. D3mit18 loci, but not at D4mit2. D2Nds3 loci. Conclusion: Screened primers D16mit4. D3mitl7.D3mit18 showing marked polymorphisms could be used in determining the strains, origin and genetic background of mice, and these mice tested meet the reguest of inbred strain.
