摘要
目的 了解罗汉果组培苗生根所需的最佳条件。方法 在1/2MS的培养基中分别附加植物激素IAA、IBA、NAA0.1mg/L后,接入罗汉果无根小苗,置于不同温度条件下培养。结果以附加NAA效果最好,诱导率最高。达到90%。培养温度对诱导生根和根伸长有影响,低温(16℃~22℃)有利于诱导发根,高温(30℃)于利于根伸长;在日夜变温(16℃~30℃)的培养条件下,罗汉果组培苗发根多,根较长,生长粗壮。结论 在本试验中罗汉果组培苗以1/2MS附加NAA0.1mg·L^-1,日夜变温(16℃~30℃)培养的的条件下生根最好,诱导率高,根多、长,生长粗壮。
Objective To probe into the favourable conditions for rooting of in vitro plantlets of Siratia grosvenorii .Methods The phytohormones of IAA, IBA , NAA 0.1 mg·L^-1 were added into 1/2 MS medium respectively. The in vitro plantlets of Siratia grosvenorii without roots were planted in the 1/2 MS mediums, and grew at different levels of termperature.Results The highest induction rate 90%, was obtained in NAA treatment . The lower temperature(16℃ ~22℃ ) was beneficial for rooting, and the higher temperature(30℃ ) was advantageous for root growing. In the condition of temperature fluctuation from day to night, the roots grew longer and thicker. Conclusion In vitro, plantlets of Siratia grosvenorii have best rooting in the 1/2 MS medium with NAA 0.1 mg·L^-1 at the day-night temperature ranging from 16℃ to 30℃.
