Effects of Cry1Ie Protein Expressed in E coli. on Biochemical Indicators and Flora of Intestinal Tract in Mice

摘要 为检测大肠杆菌表达的Cry1Ie蛋白为模式蛋白对小鼠血液生化和肠道菌群的影响,提供Cry1Ie基因的食用安全依据。将72只ICR小鼠分为Cry1Ie基因表达蛋白组(以833μgkgBWd-1剂量喂饲小鼠30d)、小牛血清白蛋白对照组和空载体大肠杆菌菌液对照组,观察小鼠体重、测定血液生化指标(谷丙转氨酶、谷草转氨酶、总蛋白、血清白蛋白、尿素氮、肌苷、甘油三酯、胆固醇、血糖)和肠道菌群数量(肠杆菌、肠球菌、双歧杆菌、乳酸杆菌)。Cry1Ie基因表达蛋白组与大肠杆菌菌液对照组、小牛血清白蛋白对照组比较,小鼠体重、血液生化指标和肠道菌群各指标的影响均未发现差异有显著性。实验结果显示在本实验条件下,Cry1Ie基因表达蛋白未对动物健康产生影响。 Crylle expressed protein is used as a model protein to examine its effects on biochemical indicators and flora of intestinal tract in mice. 72 ICR mice were randomly divided into one experimental group and two control groups. The experimental group was treated with 833 μg/kg·BW·d^-1 Crylle expressed protein. The two control groups were treated with solution of bovine serum albumin and culture solution of E coli. with control plasmid without Crylle, respectively. After 30 days, body weight, biochemical indicators (AST, ALT, TP, ALB, BUN, CRE, TRIG, CHOL, GLU) and numbers of flora of intestinal tract (enteric bacilli, enterococci, Bacillus bifidus, lactobacilli ) were examined. There were no significant differences between experimental group and control groups in body weight, biochemical indicators and numbers of flora of intestinal tract. In the present study, the Crylle expressed protein had no toxicity to experimental animals. These results offer basic information for safety evaluation of new transgenic Crylle expressed food, and establish foundation for food safety assessment of exogenous proteins derived from modem biological technique.
