香烟烟雾的氧化应激效应及褪黑素的干预作用

摘要 目的通过体外实验了解香烟烟雾作用下细胞的氧化应激状况,并了解褪黑素对这种状况的影响。方法选择(200±20)g雄性Wistar大鼠,无菌采血并分离淋巴细胞。用pH值7·2的磷酸盐缓冲液(PBS)作为吸收液制备0·2支/毫升的水溶性香烟烟雾溶液(CSS),并以终浓度8×10-3支/毫升的香烟烟雾溶液作用于细胞。前期试验中用0·05和0·1mmol/L褪黑素预处理香烟烟雾作用的细胞,发现0·1mmol/L的褪黑素增加细胞活力的作用比0·05mmol/L强,因此作为后续试验的干预浓度。实验中以RPMI1640为培养液,在37℃、体积分数5%CO2的条件下孵育细胞。实验设对照组、褪黑素作用组(MLT组)、香烟作用组(CSS组)、褪黑素与香烟共同作用组(MLT+CSS组)。用0·1mmol/LMLT对MLT和MLT+CSS组细胞预处理2h后,在CSS和MLT+CSS组细胞中加入香烟烟雾溶液,再孵育2h。用AlamarBlue摄入法检测细胞活力,彗星试验检测DNA损伤,Nitrite法测定SOD活性,TBA荧光法测定脂质过氧化水平(LPO),考马斯亮蓝G250测定蛋白质水平以校正SOD及LPO值。结果CSS组彗星尾长以及LPO水平均高于对照组,SOD活性低于对照组,差异显著。与CSS组相比,MLT+CSS组细胞活力增加,DNA损伤减轻,SOD活性增强,LPO水平降低,差异显著。结论体外实验中,8×10-3支/毫升的香烟烟雾溶液处理细胞出现氧化应激反应。0·1mmol/LMLT能增强细胞活力,增加SOD活性,减轻脂质过氧化和DNA损伤,有效拮抗CSS的氧化损伤作用。
