运用全自动酶免分析系统测定缺血修饰白蛋白 被引量:8

摘要 目的建立白蛋白结合钴试验方法检测缺血修饰白蛋白(Ischemia Modified Albumin,IMA)。方法运用全自动酶免分析系统和微孔板,参照Bar-Or钴结合试验测定IMA,在450 nm比色,结果以吸光度单位(ABSU/ml)报告;同时用500、250、125、62.5、31.3、15.6、7.8、3.9、2.0、1.0 mmol/L的氯化亚钴代替钴试剂测定白蛋白与Co2+量的关系。结果本方法批内变异:-x=37.1 ABSU/ml,S=2.05,CV=5.52%(n=10);批间变异:-x=34.4 ABSU/ml,S=2.63,CV=7.64%(n=10)。55例健康对照组和53例胸痛病人血清IMA分别为(43.9±7.14)ABSU/ml和(57.2±24.8)ABSU/ml(P<0.01);其诊断急性冠状动脉综合征的灵敏度为81%。白蛋白与Co2+量的关系试验表明系列大量游离浓度Co2+在DTT存在时,钴白蛋白复合物的颜色并不随游离Co2+浓度增加而增加。结论采用全自动酶免分析系统建立测定IMA的方法,它方便、简单,可以大批量测定,IMA是评价心肌缺血有用的诊断工具,但白蛋白结合钴试验的显色机制有待进一步探讨。
