血栓调节蛋白和纤溶酶原激活物抑制剂与糖尿病心肌病变的相关关系 被引量:1

Role of thrombomodulin and plasminogen activator inhibitor-1 in diabetic cardiomyopathy

摘要 目的探讨血浆血栓调节蛋白(TM)和纤溶酶原激活物抑制剂(PAI)-1在糖尿病心肌病变中的作用及相互关系。方法Wistar大鼠60只随机分为实验组(36只)和对照组(24只),实验组一次性腹腔注射链脲佐菌素(STZ),对照组腹腔注射枸椽酸缓冲液。分别于注射后4、8、12周时每组各处死8只取血测TM和PAI-1,并同时取心脏组织制片,在光镜和透射电镜下观察心肌的病理改变。结果实验组血浆TM和PAI-1均在第4周时较正常对照组上升(P<0.05),在8、10周时呈持续上升趋势(P<0.01),实验组血浆TM和PAI-1,呈正相关(r=0.786,P<0.01),对照组的血浆TM和PAI-1无相关。心肌超微结构在第4周出现病理改变,并随着时间延长,8、12周更加明显。结论血浆TM和PAI-1升高在糖尿病心肌病中起着重要作用。 Objective To explore the role and relationship of thrombomodulin and plasminogen activator inhibitor-1 in diabetic cardiomyopathy. Methods Sixty Wistar rats were divided randomly into two groups.There were twenty four in normal control group (NC) and thirty six in diabetes mellitus group (DM).The subjects in DM group were injected intraperitoneally with streptozotocin while the ones in NC group were injected intraperitoneally with citric acid buffer.At 4,8,12 weeks after injection,there were 8 rats in every groups respectively killed.The blood samples were obtained to measure the levels of TM and PAI-1.The heart samples were examined using light microscope and electronic microscope. Result The levels of TM and PAI-1 of DM were higher than that of NC at every stages (P〈 0.05),the changes of myocardium ultrastructrue were obvious,these changes were more significant at twelve weeds. Conclusion Higher plasma levels of TM and PAl-1 plays an important role in diabetic cardiomyopathy.
