三七三醇皂苷配合抗高血压治疗预防高血压性脑卒中 被引量:6

Combined Antihypertensive and Panaxtriol Saponins Therapy Prevents Stroke in Stroke-prone Renovascular Hypertensiven rats

摘要 目的探讨三七三醇皂苷对高血压基础上自发性卒中的预防效果。方法采用易卒中型肾血管性高血压大鼠(RHRSP)120只,随机分为4组,每组30只,肾动脉狭窄术后4周始,分别用三七三醇皂苷、福辛普利、福辛普利+三七三醇皂苷或生理盐水灌胃1次/d,持续10周,每2周测动脉血压1次,并以30只正常血压大鼠做对照。记录出现卒中的大鼠数,肾动脉狭窄术后14周处死所有大鼠,病理检查各组动物脑卒中发生病灶数和面积,并进行比较分析。结果服药2周后,福辛普利、福辛普利+三七三醇皂苷组的血压水平低于生理盐水组。用药10周期间,三七三醇皂苷、福辛普利、福辛普利+三七三醇皂苷和生理盐水组大鼠卒中发生率分别为23·3%、13·3%、10%和50%,各治疗组卒中发生率与生理盐水组均有显著差异(P<0·05);各组卒中病灶平均面积分别为(1888·6±912·5)、(2578·7±1873·9)、(1633·6±1025·1)和(3802·6±1221·1)μm2,其中三七三醇皂苷组和福辛普利+三七三醇皂苷组与生理盐水组有显著差异(P<0·05)。结论三七三醇皂苷或血管紧张素转换酶抑制剂均可降低易卒中型肾血管性高血压大鼠的脑卒中发生率,两药合用还能减轻卒中的严重程度。 Objective To evaluate the effects Panaxtriol Saponins for preventing spontaneous stroke in renovasculuar hypertensive rats. Methods One hundred and twenty stroke-prone renovascular hypertensive rats(RHRSP) were divided randomly into 4 groups with 30 animals in each group: group A( Panaxtriol saponins, PTS), group B( Fosinopril), group C[-Panaxtriol saponins [PTSI + Fosinopril] and group D (Sodium normal solution). Medications were given 4 weeks after operation by garage for 10 weeks. SBP was measured by indirect tail-cuff sphygmomanometer every 2 weeks. Thirty mormotensive rats were used as controls. We also used 30 non-hypertensive SD rats as negative control (group E). All animals were euthanized at 14th weeks. Sizes and numbers of stroke foci were counted. Results Blood pressure in group B and C was lower than that in group D(P〈0.05 respectively). The incidence of spontaneous stroke in group A, group B and group C was 23.3%, 13.3% and 10%, which was significantly lower than that in group D( 50%, P〈0.05 respectively). The average area of stroke focus in group A and group C (was 1888. 6 ± 912.5 and 1633.6 ± 1025. 1 μm^2 ), which was satistically significant smaller than that in group D(3802.6 ±1221.1μm^2) ( P〈0.05 respectively). Conclusion Combined panaxtriol saponins (PTS) and fosinopril treatment decrease the incidence and severity of stroke in experimental hypertensive rats.
