联合微量营养素对胰岛β细胞保护作用的图像分析研究

摘要 目的研究微量营养素联合生物学效应对胰岛细胞保护作用的特点,为有效防治糖尿病提供科学依据。方法应用MLDS(multiple low dosage of streptozotocin)方法制备小鼠T1DM(1型糖尿病)模型,应用免疫组织化学方法进行胰岛β细胞胰岛素染色,通过计算机图像分析方法评价胰岛β细胞胰岛素合成功能。结果微量营养素添加组小鼠血糖水平明显低于糖尿病模型组;胰岛平均灰度值明显低于糖尿病模型组(P<0.01);Insulin阳性表达区域面积与胰岛面积比明显高于糖尿病模型组(P<0.01)。结论微量营养素不同种类与剂量组合可保护胰岛素合成功能,改善胰岛功能状态与糖代谢。 Objective To investigate the biological roles of united micronutrients in βceUs protection of pancreatic islets from diabetes to prevention and treatment diabetes. Methods The injection of muhiple low dosage of streptozotocin was used to induce mice TIDM model The β ceils of islets in pancreas sections was stained by immunohistochemical method and the staining resuhs was measured by computer image analysis method. Results Both the blood glucose level and the mean gray value of groups supplemented with united micronutrients was lower than that of T1DM model group, but insulin staining positive area of mices from micronutrients-supplemented groups was higher than that of T1DM model group. Conclusion The united micronutrients, with suitable dosage and combination, improved islets function of T1DM mices.
