The relationship between heat shock protein 70 and apoptosis of astroctoma

摘要 目的:研究热休克蛋白70(heat shock prote in 70,HSP70)与星形细胞瘤凋亡的相关性。探讨其对肿瘤细胞生长和增殖的影响。方法:采用组织化学法,末端标记法,原位杂交等技术对85例星形细胞瘤的HSP70的表达及其与肿瘤凋亡的相关性进行研究。结果:HSP70表达在正常脑组织与星形细胞瘤组织之间有显著差异(P<0.05)星形细胞瘤中HSP70表达的免疫组化与原杂交较结果相符。肿瘤不同分级中Ⅰ、Ⅱ级与Ⅲ、Ⅳ级之间凋亡指数(AI)存在显著差异(P<0.05)。结论:HSP70在星形细胞瘤的表达与凋亡指数(AI)呈负相关。HSP70的表达强度随肿瘤分化程度的降低而降低,与肿瘤分化程度呈负相关。 Objective:To evaluate the correlation between heat shock protein 70 (HSPT0)and apoptosis of astrocytoma and the effects of HSP70 on the growth and proliferation of astrocytoma. Methods. Eighty five cases of astrocytoma were studied by TdT-mediated dUTP nick end labeling, immunohis tochemistry, in situ hybridization. The expression of HSP70 in normal brain tissues and astrocytoma tissues were compared. The correlation of HSP70 expression and apoptosis of astrocytoma tissue were analyzed. Results. Apoptosis in astrocytoma was different with differentiation of the neoplasm. Among the cases of low grade (gradeⅠ and Ⅱ ) astrocytoma and high grade (grade Ⅲ and Ⅳ ) astrocytoma, the lower grade ( grade Ⅰ and Ⅱ ) astrocytoma, manifested the higher apoptosis index (AI) (P〈0.05 ), It suggested that apoptosis in astrocytoma decreases gradually in association with the differentiation of neoplasm. The results of in situ hybridization and immunohistochemistry of HSP70 in astrocytoma were correspondent, there was negative correlation between AI and HSP70 in astrocytoma, which suggested that HSP70 depresses the apoptosis in astrocytoma. Conclusion: AI and expression of HSP70 are related to the differentiation of astrocytoma, which is of value to distinguish different malignancies of astrocytoma.
