摘要
6一8周龄雌性Lewis大鼠,在用视网膜抽提物(RE)或S-Ag加弗氏完全佐剂(CFA)免疫制成实验性自身免疫性色素膜网膜炎(EAU)动物模型之前鼻腔内给予相同抗原每天1次,共10天。结果表明:①该耐受疗法可有效地抑制临床疾病的发生率和严重程度;②耐受组疾病的抑制与不同淋巴器官中γ干扰素(IFN-γ)分泌细胞数的减少和RE特异性皮肤迟发性过敏反应(DTH)的抑制相一致;③病程中血清抗体的变化与对照组差异无显著性。提示鼻腔给予RE或S-Ag抑制EAU与其诱导的致病性T淋巴细胞的抑制有关。
ewis rats,aged 6-8 weeks,were given nasally with retinal extract (RE)orS-Ag daily for 10 days prior to immunization with same antigens emulsified in completeFreund’s adjuvant(CFA).The results sho wed that both the severity and the incidenee of theclinical responses of experimental autoimmune uveoretinitis(EAU)were effectively sup-pressed. Meanwhile, the suppression of EAU was consistent with the inhibition of RE orS-Ag reactive interferon-γsecreting cells in some lymphoid organs and with the decreaseof skin delayed-type hypersensitivity responses to RE. But rats which were tolerized withRE or S-Ag exhibited a similar antibody response when compared to controls(phosphate-buffered saline given nasally).These results demonstrated that nasal inductionof tolerance suppressed the onset and severity of EAU by inhibiting the pathogenic Tlymphocytes.
