摘要
为探讨内皮源性舒血管因子(endothelium-derivedrelaxingfactor,EDRF)在急性肾功能衰竭(ARF)发生发展中的作用及其机制,在甘油致大鼠ARF模型上,分别观察了NO前体L-arginine(L-Arg)和NO合成酶抑制剂N-nitro-L-arginine(L-NNA)的作用。实验结果表明:ARF大鼠NO合成减少,L-Arg能明显降低ARF大鼠血清中的BUN和Scr,尿中NAG和肾皮质中的钙含量,而提高血清NO水平,增加肾组织cGMP含量,改善肾功能;L-NNA则使ARF恶化。提示:EDRF在甘油所致ARF大鼠的发生发展中可能起重要的保护作用。
The possible role of endothelium-derived relaxing factor(EDRF),identifiedas nitric oxide (NO),in the patho genes is of glycearol-induced acute renal failure(ARF)wasexannned in rats.The effects of L-arginine(L-Arg),the physiological No precursor,and NO synthesis inliibitor,NG-nitro-L-argining(L-NNA)were observed on a model ofglyeerol-induced ARF, respectivily.Male Sprague-Dewley rats depleted with water for16 h were used to indueed ARF by intramuscular injection of glycerol.The rats were dividedinto four groups;ARF group(ARF),Glycerol+L-Arg group(GLA),Glycerol+L-NNAgroup(GLN) and control group(CON).It was found that the values of BUN(bloodurea nitrogen) and Scr(Serum creatinine) in ARF and GLN groups were increasedmore than those in GLA group. The activity of UNAG,and the contents of renalco rtex calcium and renal cAMP were significantly increased in both ARF and GLNgroups as compared with those of GLA,but the contents of serum NO and renalcGMP in both ARF and GLN groups were much lower than those of GLA. TheseresuLts indicate that the L-Arg increases the contents of serum NO and renalcGMP, alleviates renal cortex calcium over-load and improves renal function,WhileL-NNA exaggerates the impaired renal founction. It is therefore concluded thatEDRF may play an important protective role in the pathogenesis ofglycerol-induced ARF.
