实时荧光PCR结合探针熔解曲线检测亚甲基四氢叶酸还原酶C677T基因多态性及其与肝硬化的相关性研究

摘要 聚合酶链反应.限制性片段长度多态性(PCR-RFLP)是当前常用的检测基因多态性的方法之一,该法不仅操作时间长,且在采用限制性内切酶对PCR产物进行酶切后,通常采用溴乙锭或银染的方法观察结果。溴乙锭严重危害人体健康,而银染试剂相当不稳定。因此有必要建立一种快速且对人体危害性较低的多态性检测方法。实时荧光聚合酶链反应(PCR)是一种将先进的光学技术、温控技术、荧光素标记技术以及计算机软件有机结合的新型PCR,能够对PCR每一个循环的荧光强度进行监测,故称为实时荧光PCR。双探针杂交技术是一种重要的实时荧光PCR技术,通常用于基因的定量检测,但是如果结合探针熔解曲线则可用于基因多态性和突变的检测。
