骨髓间充质干细胞在鸡蛋膜为支架材料上复合培养的实验研究 被引量:7

摘要 目的探讨鸡蛋膜作为组织工程支架材料同大鼠骨髓间充质干细胞(bonemarrowmesenchymalstemcells,BMSCs)复合培养的可行性。方法利用材料试验机对鸡蛋膜进行力学性能评价;用浓度0.1mol/LHCl预制鸡蛋膜支架材料且复合Ⅰ型胶原后,采用梯度离心法分离大鼠骨髓间充质干细胞;将第2代的BMSCs同鸡蛋膜支架材料复合培养,连续培养7、14d后分别用扫描电镜观察。结果鸡蛋膜具有一定的力学强度;细胞同鸡蛋膜复合培养后容易贴附生长,7d后鸡蛋膜表面有大量的细胞生长,14d后细胞连接成片且鸡蛋膜网状孔隙中有大量的细胞生长。结论鸡蛋膜具有良好的力学特性,同时又具有空间网状结构,适合细胞生长、增殖,是作为组织工程的理想支架材料之一。 Objective To study the feasibility of eggshell membrane as scaffolds loading with rat bone marrow mesenchymal stem cells (BMSCs). Methods The mechanical characteristic of eggshell membrane was estimated by material experimental instrument. The eggshell membrane scaffold was prepared with 0.1 mol/L HC1 and complexed with type Ⅰ collagen. The BMSCs were isolated from rat bone marrow with density gradient centrifugation,then the second generation BMSCs were co-cultured with eggshell membrane scaffold, and observed under scanning electron microscope at 7 days and 14 days after continuous cultivation. Results The eggshell membrane had a certain mechanical intensity. The BMSCs might easily adhere and growth on eggshell membrane after co-cultivation massive BMSCs growth on eggshell membrane surface were found at 7 days,the cells proliferated into patches and filled in the net pores of eggshell membrane scaffold at 14 days after cultivation. Conclusion The eggshell membrane has either favorable mechanical characteristic or spatial network structure. It is appropriate for cell growth and proliferation. Thus the eggshell membrane is one of the ideal scaffolds for tissue engineering.
