摘要
目的应用双向电泳(two-dimensional electrophoresis,2-DE)技术,对比年龄匹配的不同性别核型先天性白内障蛋白质2.DE的表达,寻找差异的蛋白质。方法收集年龄匹配的核型先天性白内障患儿的混浊晶状体组织,采用固相pH梯度(IPG)等点聚焦2-DE技术进行蛋白质的分离,使用Image Master图像分析软件分析电泳图像,对比蛋白质表达的差异。结果大部分晶状体蛋白质分布在Mr14400~66000、等电点(PI)5~9的范围内,呈高丰度表达的蛋白质斑点分布在Mr20100~45000、等电点(PI)6~8的区域内。经软件分析,女性核性先天性白内障患儿识别出(452±23)个蛋白斑点,男性患儿组识别出(421±19)个蛋白斑点,将不同性别患儿的2.D结果进行匹配比较。发现有119个斑点的蛋白质表达量有变化:其中14个点蛋白质表达量减少200%以上,编码19号的蛋白质表达量减少了500%以上;其中7个蛋白斑点表达量增加了200%以上,编码384号的蛋白斑点增加350%以上。结论性别不同的核性先天性白内障患儿的蛋白质表达存在差异。
Objective To investigate the different expressed proteins in the different gender of congenital cataract with the matched years old by twodimensional electrophoresis(2-DE). Methods The congenital nuclear cataract lens with matched years old were collected. The separation of lens proteins was made by immobilized pH gradients 2-DE. The different expressed proteins were analyzed by 2-DE imaging analyzer. Results The maps of 2-DE showed that major lens proteins were in the section of PI 5-9 with relative molecular weight of 14400 - 66000 , while more abundant lens proteins were in the section of PI 6- 8 with relative molecular weight of 20100- 45000. About (452 ± 23) prtein spots were detected by image masteranalysis software in female congenital nuclear cataract group and (421 ± 19) protein spots in male group. There were 119 protein spots different expression between two group. The expression of male lens protein compared with female decreased to more than 200% in 14 spots, in which spot No. 19 decreased to 500%. The expression of lens protein increased to more than 200% in 7 spots, in which spot No. 384 increased to 350%. Conclusion The expressed proteins in the congenital nuclear cataract lens are different between male and female with the roached old.
