摘要 目的观察转化生长因子β1(transforminggrowthfactor-β1,TGF-β1)对体外培养的牛角膜内皮细胞细胞外基质(extracellularmatrix,ECM)合成的影响。方法将体外培养的牛角膜内皮细胞分为实验组和对照组,经0μg·L-1(对照)、0.1μg·L-1、1.0μg·L-1、10.0μg·L-1、100.0μg·L-1浓度的TGF-β1处理48h后,用SABC免疫组化法分别检测角膜内皮细胞纤维连接蛋白和层粘连蛋白的合成情况。结果与对照组相比,一定浓度(1.0μg·L-1及10.0μg·L-1)的TGF-β1能明显促进体外培养的角膜内皮细胞纤维连接蛋白及层粘连蛋白的合成,且呈剂量依赖性。但浓度为0.1μg·L-1及100.0μg·L-1的TGF-β1组OD值与对照组比较,差异无显著性(P>0.05)。结论一定浓度(1·0μg·L-1及10.0μg·L-1)TGF-β1能促进牛角膜内皮细胞ECM的合成,提示TGF-β1可能在角膜内皮细胞损伤修复中扮演重要角色。 Objective To investigate the effect of transforming growth factor-β1 ( TGF-β1 ) on extracellular matrix (ECM) synthesis in cultured bovine corneal endothelial cells. Methods Cultured 3- 5 passage bovine corneal endothelial cells were divided into control group and experimental group. After being treated with 0 μg·L^-1 (control) ,0.1 μg·L^-1,1.0 μg·L^-1,10.0 μg·L^-1,100.0 μg·L^-1 TGF-β1 for 48 hours, laminin(LN) and fibronectin(FN) synthesis in bovine corneal endothelial cells were examined respectively by immunohistochemical technique. Results In comparison with control group, TGF-β1 significantly promoted the sythesis of LN and FN at 1.0 μg·L^-1 and 10.0 μg·L^-1(p〈 0.01), and TGF-β1 enhanced the production of LN and FN in the endothelial cells in a dose-dependent manner, but remained at normal level in those at 0. 1μg·L^-11 and 100.0 μg·L^-1(P〉0.05).Conclusion At the concentrations of 1.0 μg·L^-1 and 10.0 μg·L^-1, TGF-β1 can increase ECM synthesis in bovine corneal endothelial cells, the data suggest that TGF-β1may play an important role in corneal endothelial healing process.
