神经生长因子β对兔实验性青光眼视网膜神经节细胞的保护作用 被引量:3

摘要 目的探讨神经生长因子β(NGFβ)对青光眼性视网膜神经节细胞(RGCs)损害的保护作用。方法建立家兔青光眼模型,并动态观察眼压。于术后14d、21d随机选择一眼玻璃体腔注射NGFβ1μg和细胞色素C0.1mg(实验组),对侧眼仅注射细胞色素C0.1mg作对照(对照组),共10只兔。术后28d处死实验兔摘取眼球及视神经作组织学观察,并用核苷酸末端转移酶介导的dUTP缺口末端标记法(TUNEL)检测RGCs凋亡状况。结果组织学检查显示术后28d时视神经神经纤维丢失程度实验组有8例轻于对照组,1例相似,1例较对照组为重。对照组视神经节细胞层凋亡阳性率13.46%,明显高于实验组的10.60%(P<0.01)。结论NGFβ对青光眼性视网膜神经节细胞损害具有一定的保护作用。 Objective To investigate the protective effect of nerve growth factor-β(NGF-β) on the survival of retinal ganglion cells(RGCs) in the experimental glaucoma rabbits. Methods The experimental glaucoma was produced in rabbits and intraocular pressure was measured on the 14 th and 21st day. A combination of NGF-β(1μg) and cytchrome C (0. 1 rag) was injected into vitreous body of one eye randomly (treated group). The other eyes of rabbits received cytchrome C (0. 1 rag) only served as control group. On the 28 th day, the eye global with optic nerve was enucleated for histologic examination. TUNEL method detecting apoptic cells was taken. Results Eight eyes in treated group were lessserious, 1 eye more serious than that those in control group. The positive rate of apoptosis in treated group was 10. 60% ,which was significantly less than that in control group(13.46%) (P〈0. 01). Conclusion NGF-β could protect the RGCs from damage in glaucoma.
