氨茶碱对体外不同密度嗜酸细胞凋亡和白介素5受体α mRNA表达的影响 被引量:2

摘要 目的观察氨茶碱(抗哮喘药)对哮喘豚鼠体外不同密度嗜酸细胞 (Eos)凋亡及白介素5受体α(IL-5Rα)mRNA表达的影响。方法制备卵蛋白激发哮喘豚鼠动物模型,48 h后,行支气管肺泡灌洗,分离低密度及正常密度Eos(HEos,NEos),HEos及NEos分别与(1×10-6-1×10-3mol· L-1)氨茶碱共培养24 h;以原位杂交方法检测不同Eos的IL-5Rα mRNA表达,在3’末端脱氧核苷转移酶介导的脱氧三磷酸尿苷缺口末端,标记法检测细胞凋亡。结果氨茶碱干预24 h后,可见Eos细胞凋亡增加,不同密度 Eos IL-5Rα mRNA表达也增加,且与氨茶碱浓度呈剂量依赖关系,IL-5Rα表达与Eos凋亡呈正相关(P<0.05)。结论氨茶碱可促进不同密度Eos表达IL-5Rα,抑制其细胞因子活动,促进Eos凋亡。 Objective To investigate the effects of aminophylline on expression of interleukin - 5 receptor α ( IL - 5 Rα) mRNA and apoptosis in eosinophils (Eos) of bronchalveolar lavage fluid (BALF) in asthmatic guinea pig. Methods After guineas pigs were stimulated by ovalbumin 48 hours, hypodense Eos(HEos) and normodense Eos(NEos) were purified from BALF by gradients of percoll. Eos cultured in PRMI 1640 medium. Add aminophylline ( 1×10^-6 ~1×10^-3 mol·L^- 1 ) in the wells. The mRNA of IL-5Rα in Eos was measured by hybridization. Apoptosis was detected by TdT - mediated dUTP nick end labeling. Results Eos cultured under the condition of aminophylline administration in vitro, apoptotic Eos increased, IL -5 Rα mRNA expression increased in a dose -dependent manner. There was significant correlation between Eos apoptosis and IL- 5Rα mRNA expression. Conclusion Aminophylline promote Eos apoptosis, increase expression of IL - 5 Rα in dose - dependent manner in vitro.
