摘要
目的观察血管内皮细胞受到氧自由基损伤后,血管内皮细胞粘附分子-1(VCAM-1)、血小板衍生生长因子B链(PDGF-BB)表达、内皮素(ET)以及血管紧张素Ⅱ(AⅡ)分泌的改变,研究扇贝裙边提取物-糖胺聚糖(SS-GAG)对上述变化的影响,探讨SS-GAG对血管内皮细胞损伤的保护作用。方法体外培养人脐静脉内皮细胞(HUVEC),建立氧自由基诱导的HUVEC损伤模型,采用ELISA法观察SS-GAG对Fenton体系所引发的氧化损伤后HUVEC表达VCAM-1,PDGF-BB的影响;应用放射免疫方法,观察SS-GAG对氧化损伤的HUVEC分泌ET及AⅡ的影响。结果与对照组比较,Fenton体系损伤组血管内皮细胞(VEC)VCAM-1,PDGF-BB的表达明显提高(P<0.01),ET及AⅡ的分泌明显增多(P<0.01)。而细胞经不同浓度的SS-GAG预先处理后,上述表达及分泌均减少(P<0.01)。结论SS-GAG可减少氧自由基损伤血管内皮细胞后VCAM-1,PDGF-BB的表达以及ET和AⅡ的分泌,提示SS-GAG具有保护血管内皮细胞的作用,此作用可能是其抗AS的机制之一。
OBJECTIVE To observe the changes of vascular endothehum damaged by oxygen free radical , including the experession of vascular endothelial cell adhesion molecule-1 (VCAM-1 )and platelet derived growth factor B chain(PDGF-BB), the secretion of endothelin and angiotensin Ⅱ ,and to investigate the effects of the scallop skirt-glycosaminoglycan (SS-GAG) on the endothelial cell damage by oxygen free radical. METHODS The endothelial cells of human umbilical vein (HUVEC)were cultured, and the Fenton reaction oxygen free radical damage model of endothelial cells was established. After the damage of HUVEC by Fenton reaction. The experession of vascular endothelial cell adhesion molecule-1 (VCAM-1)and platelet derived growth factor B chain(PDGF-BB)were determined by the methods of ELISA. The secretions of endothelln and angiotensin Ⅱ were measured by radio-immunity assay.RESULTS Compared with the control group, the experessions of vascular endothelial cell adhesion molecule-1(VCAM-1)and platelet derived growth factor B chain(PDGF-BB), the excretions of endothelins and angiotensin Ⅱ in HUVEC were increaced obviously( P 〈 0.01 ) in Fenton reaction damage group. Meanwhile, those experession and the excretion were decreased obviously( P 〈 0.01) in the groups pre-treated by SS-GAG. CONCLUSION SS-GAG can inhibit the secretion of endothelins and angiotensin Ⅱ , the experession of vascular endothelial cell adhesion molecule-1 (VCAM-1 )and platelet derived growth factor B chain(PDGF-BB)in HUVEC after the damage of oxygen free radical.The anti-atherosclerotic mechanism of SS-GAG may relate to it's protection on the endothelial cell.
