大鼠外周炎症组织内β-内啡肽的表达 被引量:3

摘要 目的:研究大鼠外周炎症组织中的β-内啡肽在炎症过程中表达的变化,为进一步探讨内源性阿片肽在外周炎症性疼痛中所起的作用。方法:在大鼠后足注射1%角叉菜胶产生外周炎症。分别在正常和炎症后6、12、24、72h取大鼠后足皮下组织,然后:①用HE染色后,观察炎症组织内的炎性细胞变化;②用免疫组织化学的方法检测大鼠后足皮下组织的β-内啡肽,并用图像分析系统进行半定量分析。结果:①在炎症过程,皮下组织中可见大量的白细胞浸润。②正常皮下组织无β-内啡肽表达。炎症6h大鼠足底皮下组织可见β-内啡肽表达,表达量在炎症24h到达最高峰,72h下降至6h水平。β-内啡肽免疫阳性的白细胞数在6、12、24h没有明显差异(P>0·05),72h后降低。结论:大鼠外周炎症组织中的β-内啡肽来源于浸润到炎症组织的白细胞,β-内啡肽表达量及含β-内啡肽免疫阳性的白细胞数随炎症过程变化而变化。 Objective:To observes source and changes of β-endorphine at peripheral inflamed tissue of rats, which is foundation for study analgesia of endorphine at peripheral inflamed tissue. Methods: Peripheral inflammation was induced by injected carrageenan to the hindpaws of the rat. Tissue samples were harvested 6, 12, 24 h and 72 h after injection of carrageenan. Subcutaneous paws tissue was obtained from the plantar. ①the change of infiltrated inflammatory cells in the paw tissue with HE Stained was observe. ②the β- endorphine in the subcutaneous paw tissue was identified by immunohistochemistry and half quantified by image analysis system. Result: ①There was a lot of infiltrating immune cells in the subcutaneous paw tissue during inflammation.② The-endorphine was not detected in the normal subcutaneous paw tissue. There was expression of-endorphine in subcutaneous paw tissue at 6th hour after injection of carrageenan. The amount of-endorphine was the highest in subcutaneous paw tissue at 24th hour during inflammation. There was not significantly difference at 6 h and 72 h. The number of β-endorphine-producing cells was not significantly different at 6,12,24 h during inflammation( P 〈0.05). But it decreased at 72 h. Conclusion:The β- endorphine in the inflammatory tissue comes from infiltrating leukocytes. The amount of β-endorphine and β-endorphine-producing cells change with inflammation.
