哺乳动物初级精母细胞体外培养的研究

摘要 目的:探讨哺乳动物初级精母细胞体外培养分化的可能性。方法:制备雄性大鼠睾丸初级精母细胞,与非洲绿猴肾上皮细胞(Vero细胞)共培养,观察初级精母细胞体外培养过程中的生长行为,用5-溴-2-脱氧尿苷(BrdU)标记跟踪观察大鼠初级精母细胞在体外培养中的分化情况。结果:大鼠初级精母细胞与Vero细胞共培养后能进行减数分裂阶段分化,生成精子细胞;BrdU标记检测结果显示,培养前在体内标记BrdU的细胞为初级精母细胞,培养后在有BrdU标记的细胞中有早期精子细胞。结论:哺乳动物初级精母细胞在体外培养中能进行分化。
