摘要
目的探讨基质金属蛋白酶9(matrix metalloproteinases-9,MMP-9)在人创伤性脑损伤(traumatic brain injury,TBI)后挫伤皮层中的表达情况,包括表达位置、表达强度和表达时相。方法挫伤区皮层的标本来自于24位TBI患者,取样时间从伤后5h到5d,利用免疫组化技术测定MMP-9的表达强度。结果在人TBI后的挫伤区皮层中,MMP-9表达明显上调,表达高峰为伤后24-48h,MMP-9主要在神经元细胞中表达,神经胶质细胞中也有少量表达。结论MMP-9在人TBI后的挫伤区皮层中表达上调,提示其可能在TBI后的病理生理过程中起着重要作用。
Objective The aim of current study was to analyze the expression of matrix metalproteinases-9(MMP-9) in contused human brain tissue, and to identify the expressive location, quantitation and time points. Methods Contrustal brain tissue biopsies were obtained from 24 patients with surgery for brain from 5 hours to 5 days after trauma. The expression of MMP- 9 was detected by immunohistochemistry. Results The result showed an immediate, strong and persistent up-regulation of MMP-9 in the area surrounding the injured brain as early as 5 hours postinjury and was keeping at high level up to 5 days. The maximal expression of MMP-9 was detected at 24 to 48 hours post injury. Expression of MMP-9 was mainly located at neurons and slight expression in a few neuroglial cells. Conclusion The results suggest that MMP- 9 is up- regulate obviously in the early phase of human brain contusion. MMP-9 might play important roles in the pathophysiological course after TBI.
