HPLC法测定四季三黄胶囊中盐酸小檗碱的含量

Determination of Berberine Hydrochloride in Siji Sanhuang Capsules

目的对中药复方制剂四季三黄胶囊中有效成分盐酸小檗碱进行含量测定。方法采用高效液相色谱法,Ag-ilentHypersilODS色谱柱(4.6mm×150mm,5μm),乙腈-0.05mol/L磷酸二氢钠溶液(用磷酸调pH值至3)(30∶70)为流动相,检测波长为270nm,流速1.0mL/min,柱温25℃。结果盐酸小檗碱在0.094～0.470μg范围内与峰面积呈良好的线性关系,r=0.9983,平均加样回收率为98.5%(n=5),RSD=2.3%。结论该方法快速简便,灵敏度和稳定性高,可有效控制四季三黄胶囊的内在质量。

Objective To establish a method for the determination of berberine hydrochloride in Sift Sanhuang capsules （SSC）. Methods The RP - HPLC method was performed on Agilent Hypersil ODS column （4. 6 mm × 150 mm, 5μm）. Acetonitrile and 0.05 mol/L NaH2PO4 water （adjust pH to 3 with H3PO4） were used as the mobile phase （30 ： 70）, detection wavelength was at 270 nm, the flow rate was 1.0 mL/min and the column temperature was at 25℃. Results In the range of 0. 094 to 0. 470 μg, herherine hydrochloride had a good linearity with chromatograph peaks area （ r = 0. 998 3）, the average recovery ratio was 98.5 % （ n = 5）, and RSD = 2.3 %. Conclusion This method is simple, effective and with a higher sensibility and stability. The method can he used to control the quality of SSC effectively.
