摘要
目的:建立同时测定附子中3种双酯型生物碱——新乌头碱、乌头碱、次乌头碱含量的HPLC分析方法。方法:Zorbax Eclipse XDB C18柱,40mmol乙酸铵缓冲液-乙腈,梯度洗脱,流速1.0mL·min^-1,检测波长230nm。结果:3种双酯型生物碱新乌头碱、乌头碱、次乌头碱均达到了较好的分离,分别在0.0357-1.784μg(r=0.9999),0.0126-0.632μg(r=0.9997)和0.0334-1.672μg(r=0.9997)呈良好的线形关系。结论:该方法简便、快速、灵敏,可有效地用于附子的鉴别和内在质量控制。
Objective: To establish an HPLC method for the determination of three kind of diester diterpenoid alkaloids (DDAs) in Aconitum carmichaeli and its processed products namely mesaconitine, aconitine and hypaconitine. Method: A Zorbax Eclipse XDB C18 scolumn was used, and ammonium acetate solution-acetonitrile as the mobile phase, with a flow rate of 1.0 mL· min^-1 and a detection wavelength of 230 nm. Result. Mesaconitine, aconitine and hypaconitine, were separated, and the calibration curves of them were in good linearity over the range of 0.035 7-1.784 μg ( r = 0.999 9), 0.012 6-0.632μg ( r=0.999 7) and 0.033 4-1.672μg ( r=0.999 7) respectively. Conclusion: This method is simple and accurate. It can be used in the identification and quality control of A.carmichaeli.
