苦参素对人胃癌细胞系MGC-803增殖的抑制作用 被引量:17

摘要 目的:观察苦参素对人胃癌细胞系MGC803的增殖抑制作用。方法:人胃癌细胞系MGC803接种培养24h后,实验组分别加入不同浓度苦参素(0.25g/L、0.5g/L、1g/L、2g/L、4g/L),阳性对照组与阴性对照组分别加入氟尿嘧啶(2g/L)和培养液,继续培养12h,24h,36h,48h,采用MTT法测定苦参素对MGC803细胞的增殖抑制情况。判定苦参素的剂量与MGC803细胞增殖活性间的关系,计算细胞抑制率和群体倍增时间。结果:苦参素在0.25~4g/L范围内,对MGC803细胞的增殖活性具有抑制作用,与阴性对照组比较,差异有统计学意义(P<0.001),且该作用随剂量增加而增加。与阴性对照组比较,阳性对照组和实验组细胞群体倍增时间增加。结论:苦参素对人胃癌细胞系MGC803有明显的增殖抑制作用,且具有时间、剂量效应关系。 Aim: To investigate the inhibition effect of oxymatrine (OM) on the human gastric carcinoma cell line MGC-803. Methods: MGC-803 cells were cultured for 24 h and allocated into different experimental groups. OM groups were added with 0.25 g/L, 0.5 g/L, 1 g/L,2 g/L, and 4 g/L OM, and the positive control group and negative control group were added with 2 g/L 5-fluorouracil and culture fluid, respectively. After being cultured for 12 h, 24 h, 36 h, 48 h, the proliferation inhibition of MGC-803 cells was assessed by MTT assay. The relation between the dose and cell proliferation viability was analyzed. Cell inhibitory rate and the doubling time were evaluated. Results: In the range of 0.25 -4 g/L, OM had obvious inhibitive effects on the proliferation of MGC-803 cells in a dose-dependent manner compared with the negative control group (P 〈 0.00l ). Compared with negative control group, the doubling time of both positive control group and experimental groups was prolonged. Conclusion: OM has significant inhibition effects on the proliferation of MGC-803 cells in a time-dependent and dose-dependent way.
